Danggui Buxue Decoction Protects Retinal Vascular Endothelial Cells from High Glucose-Induced Apoptotic and Oxidative Injury by Decreasing Forkhead Box O4

To explore the effect and mechanism of Danggui Buxue decoction on high glucose-induced apoptotic oxidative injury in human retinal vascular endothelial cells. Human cells were treated with glucose to mimic diabetic retinopathy condition vitro. The 3-(4,5-dimethylthiazol-2- yl)-2,5-diphenyl-2H-tetrazolium bromide assay flow cytometry used investigate cell viability apoptosis. Western blot was applied detect proteins apoptosis-related markers forkhead box O4. assessed using enzyme-linked immunosorbent analysis. group showed decrease superoxide dismutase activity, as well increase apoptosis rate, reactive oxygen species level malondialdehyde content, which reversed by treatment a dose-dependent manner. Subsequently, we found that O4 elevated cells, while dose-dependently down-regulated Deficiency could reverse Moreover, inhibitory effects attenuated overexpression. may inhibit damage induced down-regulating
